Perlecan Antibody (CSI 001-71-02) in WB

Western blot was performed using Perlecan Monoclonal Antibody (A71) (Product # CSI 001-71-02) and a 469 kDa band corresponding to HSPG2 was observed across cell lines. Whole cell extracts (30 µg lysate) of HUVEC (Lane 1), HAEC (Lane 2), SH-SY5Y (Lane 3), A549 (Lane 4) were electrophoresed using NuPAGE™ 3-8% Tris-Acetate Protein Gel (Product # EA0378BOX), 10 well. Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB33001) by iBlot™ 3 Western Blot Transfer Device (Product # IB31001). The blot was probed with t... Product Specific Information

CSI 001-71-02 is highly specific for perlecan. There is no evidence for cross-reactivity with other connective tissue proteins (vitronectin, fibronectin, elastin, collagen, laminin). CSI 001-71-02 cross-reacts with human thrombospondin. Other species have not been tested.

CSI 001-71-02 has been successfully used in ELISA, immunohistochemistry and Western blotting techniques. CSI 001-71-02 can be used in immunoprecipitation, ELISA and immunostaining of frozen PLP-fixed sections of bovine and human tissues. CSI 001-71-02 can be used in affinity chromatography to separate recombinant domain I

from tissue culture fluid.

Epitope of CSI 001-71-02 is located in domain I.

NOTE: Concentration is lot-dependent and can vary from 0.85-1.15 mg/mL

Target Information

Perlecan is a major heparan-sulfate proteoglycan (HSPG) found within all basement membranes and cell surfaces. Because of its strategic location and ability to store and protect growth factors, perlecan has been strongly implicated in the control of tumor cell growth and metastatic behavior. Perlecan possesses angiogenic and growth-promoting attributes primarily by acting as a coreceptor for basic fibroblast growth factor (FGF-2). Suppression of perlecan causes substantial inhibition of neoplastic growth and neovascularization. Thus, perlecan is a potent inducer of neoplasm growth and angiogenesis in vivo and therapeutic interventions targeting this key modulator of tumor progression may improve neoplastic treatment.
